摘  要:目的:试图观察髓鞘蛋白脂质蛋白多肽(proteolipidprotein,PLP)两种不同肽段PLP193~151和PLP178~191抗原决定簇对实验性自身免疫性脑脊髓炎(experimentalautoimmuneencephalomyelitis,EAE)模型建立的影响。方法:实验于2004-02/06在解放军总医院神经病理研究室进行。应用PLP193~151和PLP178~191免疫雌性SJL/J小鼠,制作复发缓解型实验性自身免疫性脑脊髓炎(relapseremittingEAE,RR-EAE)模型,观察其神经功能评分的变化及组织形态学改变;用不同浓度的两种肽段体外刺激经两种肽段免疫后的小鼠的淋巴细胞,并观察其增殖强度。结果:两种PLP肽段免疫的小鼠发病均具有缓解-复发的特点,并出现血管鞘形成、炎性细胞浸润及脱髓鞘等特征性改变;但是,PLP178~191诱导的模型发病时间早(P<0.01),症状较重,表现多样。不同PLP肽段及其浓度间的差异有统计学意义(P=0.000)。PLP178~191在5mg/L时A值最高(2.36±0.14),而PLP139~151在50和100mg/L的A值增高有统计学意义(P=0.000)。结论:PLP两种肽段均可诱发RR-EAE模型,但二者诱发的模型各具特点,这是由其不同抗原决定簇决定的。AIM:To observe the effect of epitopes of two different proteolipid protein(PLP) peptides:PLP139 15 and PLP178 191 on inducing the models of experimental autoimmune encephalomyelitis(EAE). METHODS:The experiment was completed in the Laboratory of Neuropathology,General Hospital of Chinese PLA from February to June 2004.The models of relapse remitting EAE(RR EAE) were established by injection of PLP139 151 or PLP178 191 peptides,and the neurological signs and morphological changes of the female SJL/J mice were observed.Their lymphocytes were stimulated in vitro by the two different peptides in different concentrations respectively.At the same time, the proliferous concentrations of lymphocytes and intensity of stimulation were observed. RESULTS:Each model manifested significant neurological symptoms,signs and features of relapse and remission.Obvious phenomena of perivascular sheathing, inflammatory cell infiltration and demyelination could be found. However,EAE mice induced by PLP178 191 showed earlier onset(P< 0.01)than that induced by PLP139 151,along with various and severe clinical signs.There was statistic significance between different species and concentrations of PLP peptide(P=0.000),with the highest A value of PLP178 191 at 5 mg/L (2.36±0.14).However, A value increased significantly at 50 and 100 mg/L of PLP139 151,and there was statistic significance(P=0.000). CONCLUSION:RR EAE model can be induced by both of PLP peptides.However,the different characters of these EAE mice are owing to the different epitopes of PLP peptides.
