摘 要:目的:观察大鼠脑出血后血肿周围神经元线粒体内细胞色素C释放规律,及中药复方平肝熄风汤的干预作用。方法:实验于2003-10在湘雅医院中西医结合研究所进行。用Ⅶ胶原酶脑内立体定位注射诱导大鼠脑出血模型,免疫组织化学法检测神经元线粒体内细胞色素C的释放情况。结果:大鼠脑出血后血肿周围神经元于6h即有细胞色素C释放,出现胞浆染色的阳性细胞;1d后胞浆染色最浓,细胞形态变得不规则,阳性细胞表达最多;3d后染色变淡,阳性细胞数减少;5d后胞浆染色继续变淡,阳性细胞继续减少。治疗组与模型组比较在1,3,5d时阳性细胞减少明显(P<0.01),染色亦较模型组淡。结论:脑出血后血肿周围神经元细胞色素C于6h即开始从线粒体释放,1d达高峰后下降;平肝熄风汤能阻制细胞色素C的释放。AIM: To observe the effect of pinggan xifeng decoction (PGXFD) on the release of cytochrome C(Cyc C) from neuronal mitochondria around the hematoma after intracerebral hemorrhage(ICH) in rats and study the releasing regularity of Cyc C. METHODS: The experiment was conducted in Institute of Combined Traditional Chinese and Western Medicine of Xiangya Hospital in October 2003.Ⅶtype collagenase was injected into rats with stereotaxic technique to build the ICH model. The immunohistochemical method was used to examine the release of Cyc C from neuronl mitochondria after ICH in rats. RESULTS: Cyc C began to release in the neuron around the hematoma of rats at 6 hours after ICH, there were some positive cells stained at the cytoplasm; after 1 day, the cytoplasm was stained strongly, the cell shape became abnormal, and the number of positive cells peaked to the top; after 3 days, the staining became weak, and the positive cells decreased; after 5 days, the staining continued to weaken and the positive cells continued to decrease. The number of positive cells in the PGXFD group was obviously less than that in the model group(P< 0.01) and the staining was also weaker than that in the model group. CONCLUSION: The release of Cyc C begins at 6 hours from neuronal mitochondria around the hematoma after ICH; it reaches the peak at 1 day and then begins to decrease. PGXFD can markedly depress the release of Cyc C.
