摘  要:目的 研究乳腺癌组织和细胞株中MageA1基因的表达情况,为MageA1基因编码蛋白用于乳腺癌免疫治疗提供依据。方法 采用RT-PCR检测 33例乳腺癌组织和 4株常用乳腺癌细胞株MCF-7、Sk-Br-3、MDA-MB-435s和TM40D中MageA1基因的表达。结果 33例乳腺癌组织中 12% ( 4 /33 )Mage-A1阳性,细胞株MCF-7和Sk-Br-3中Mage-A1阳性表达,MDA-MB-435s和TM40D中MageA1基因的表达阴性。结论 不同的乳腺癌细胞表达MageA1基因有差别,MageA1蛋白可望成为乳腺癌免疫治疗的靶抗原。Objective To study the expression of Mage-A1 in breast cancer tissues and four breast cancer cell lines and explore the possibility of Mage gene encoding proteins used as target for immunotherapy in breast cancer. Methods The expression of Mage-A1 in breast cancer tissues and four breast cancer cell lines, including MCF-7、Sk-Br-3、MDA-MB-435s and TM40D, was detected through reverse transcription polymerase chain reaction(RT-PCR). Results The positive expression rate of Mage- A1 in breast cancer tissues was 12%(4/33). Mage-A1 was positive in breast cancer cell line MCF-7 and Sk-Br-3, negative in MDA-MB-435s and TM40D. Conclusion Mage-A1 genes are expressed in breast cancer, but the positive expression of Mage-A is distinct in different breast cancer cell lines. Mage gene encoding proteins are eligible for Mage-peptide-based active immunotherapy.
