摘 要:采用正交试验方法研究了不同培养基、激素种类及其浓度对上西早生柿叶片不定芽再生的影响。结果表明,氮含量减半的MS [MS (1/2N)]培养基最好,其次是1/2 MS,MS效果最差;ZT (zeatin) 浓度为4.0 mgL-1时不定芽再生率明显高于2.0 mgL-1,在1.0mgL-1ZT中培养的叶片再生率最低;0 mgL-1 IAA (indole acetic acid) 的效果好于1.0 mgL-1和2.0 mgL-1 IAA;正交试验影响因子分析结果显示,ZT的作用最大,其次是培养基和IAA;组合MS(1/2)、4.0mgL-1ZT和1.0mgL-1IAA中不定芽再生率达86%,平均不定芽数为2.2。Effects of basal mediums, hormones and their concentrations on the shoot regeneration from leaves were compared by orthogonal design trials. The results showed that modified Murashige and Skoog (MS) [MS(1/2N)] was the most optimum for the regeneration and 1/2MS was better than MS. Shoot percentage in the medium containing 4.0mgL-1 ZT was much higher than other two concentrations, of which 2.0mgL-1ZT was much better than 1.0 mgL-1ZT and shoot percentage in the 1.0mgL-1 ZT was only 4%. There were no any positive effect when supplementing IAA in the medium, and shoot percentage and average shoots per explants were dramatically decreased in the 2.0mgL-1IAA. Data in the orthogonal trials indicated that ZT was the most effective factor in the shoot regenerating of Uenishwase persimmon and basic medium was important too, but IAA had no any beneficial effects at all. In the orthogonal trial the best result was achieved in MS(1/2N) medium containing 4.0mgL-1ZT and 1.0mgL-1IAA, in which shoot percentage and average shoots per explants were 86% and 2.2, respectively.
