MC148基因局部表达延长大鼠异基因皮肤移植的存活

摘　　要：目的:构建MC148腺病毒重组体,探讨MC148基因转染对器官移植排斥反应的抑制作用。方法:从传染性软疣病毒中克隆MC148基因,构建腺病毒重组体,局部转染Lewis大鼠皮肤后移植到Wistar大鼠。检测MC148基因在转染皮肤内的表达,观察大鼠移植皮肤的生存时间及病理改变。结果:成功构建Ad-MC148腺病毒重组体,Ad-MC148 转染后移植的皮肤存活时间较对照组明显延长(p<0.01),病理检查,转染组皮肤较少有白细胞浸润。结论:MC148基因局部转染可能抑制器官移植后白细胞的浸润而发挥免疫抑制作用。Objective: To study the immunosuppressive effect of MC148 expression on allograft rejection mediated by adenovirus vector. Method: MC148 gene was cloned from molluscum contagiosum virus ( MCV) and inserted into adenovirus vector. Recombinant Ad-MC148 was then locally transfected into the Lewis rat skin that was transplanted to Wistar rat.MC148 gene expression in transfected Lewis skin was detected. And pathological changes and survival time were investigated.Results: Recombinant Ad - MC148 adenovirus vector was constructed successively. Allograft skin survival time was prolonged and there was less leukocytes infiltration pathologically in Ad- MC148 transfected group than in controll (P<0.01) .Conclusion: MC148 gene might suppress leukocyte infiltration to prolong survival time of the allograft skin.
