癌性胸、腹腔积液的蛋白质提取及静脉回输研究

摘　　要：目的:探索癌性胸、腹腔积液蛋白质的提取方法,并对自体蛋白静脉回输后的临床疗效进行观察,从而为癌 性胸、腹积液中蛋白质的再利用提供实验依据。方法:采用透析、冷凝低速离心、变速离心和抽滤等技术对癌性积液中的蛋白 质进行分离提取。结果:该方法提取的蛋白质的浓缩倍数可达9.78倍,回收率为54.75%;蛋白质分离提取前后离子K+、 Na+、Cl-浓度均无明显差异(P>0.05),而BIL、IL 2、IL 4、IFN和TNF的浓度明显增加(P<0.05),患者自体蛋白液回输后无 发热、过敏等副反应发生,同时水肿也得到一定程度的控制,除CD+4细胞百分率和CD+4/CD+8比值明显增加外(P<0.05), CD+3、CD+8细胞百分率、总蛋白及IgG浓度均无明显改变(P>0.05)。结论:通过透析、变速离心及抽滤等技术,可从癌性胸、 腹腔积液中浓缩分离出适合静脉输注的蛋白质提取液。Objective:To explore the method by which protein was isolated from malignant hydrothorax or ascites and the clinical effect of auto-protein infused in vein,which could provide experiment evidence for the further study of the utilization of protein isolated from malignant hydrothorax or ascites.Methods:The protein contained in malignant hydrothorax and ascites was isolated and abstracted by dialysis,centrifuge and filtration.Results:The inspissation multiple of protein in the isolated and distilled course was 9.78 times,the rate of protein recovery is 54.75%:The ion concentrations of K +、Na +、Cl - have not distinct difference during the protein was isolated(P>0.05),but the concentration of BIL,IL-1,IL-4,IFN,TNF increased greatly(P<0.05).After the infusion of auto-protein,except CD 4 + and the rat of CD 4 +/CD 8 + increased(P<0.05),the concentrations of TB and IgG had not been changed,along with the percentage of CD 3 and CD 8 positive cells(P>0.05).Conclusion:Through the technique of dialysis,centrifuge and filtration,protein suit to infuse in vein could be isolated.
