摘 要:目的:观察多孔硅胶支架材料经不同化学处理后植入兔眼角膜内的生物学反应。方法:选用20只新西兰大白兔,平均分为4组,将多孔硅胶支架材料制成直径4mm的圆盘状,化学处理后植入兔角膜板层囊袋内。材料经酸溶液处理10min为Ⅰ组;酸溶液处理15min为Ⅱ组;H2O2溶液处理15min为Ⅲ组;未处理即对照组。术后每天裂隙灯观察,持续84d后取角膜行光镜、扫描电镜及免疫组织化学分析。结果:材料经化学处理后,眼睑充血减轻、分泌物减少,新生血管出现延迟,范围较小。光镜HE染色及扫描电镜观察见化学处理组多孔硅胶支架内角膜成纤维细胞和胶原蛋白较未处理组多。免疫组化染色见处理组Ⅰ型胶原沉积明显多于对照组。结论:化学溶液对多孔硅胶的表面处理可明显减轻兔角膜炎症反应,增加材料的组织相容性。Objective: To study the biological response of cornea to porous silica gel treated with different chemical agents in vivo. Methods: The porous silica gel was cut into disc with a 4mm diameter and treated with chemical agents. Twenty New Zealand white rabbits were chosen and divided into 4 groups. First group was implanted the silicon discs (4mm in diameter) treated with mixed acid solution containing 23.2% H2SO4 and 0.8% K2Cr2O7 for 10 minutes,the second group was implanted the discs treated with mixed acid solution for 15 minutes and the third group was implanted the discs treated with 30% H2O2 solution for 15 minutes. Untreated silica served as control. Discs were inserted into interlamellar stromal pockets and followed up daily with slit lamp a period of 84 days. After 3 months of surgery,corneas were excised; fibroplasias and the distribution of matrix proteins were evaluated with light microscope,scaning eleclon microscope,and immunohistochemical analysis. Results: The treated discs made eyelid congestion,conjunctival discharge,and the extent of neovascularization obviously decreased. Fibroblasia and collagen deposition in the treated discs were detected more than those in control. Type I collagen deposition in treated discs was obviously more than that in control in immunohistochemical analysis. Conclusion: Chemical treatment on silica gel is effect on decreasing rabbit corneal inflammation and enhancing tissue compatibility.
