摘 要:目的 探讨γ 干扰素(IFN- γ)、转化生长因子β3(TGFβ3) 对体外培养的创面成纤维细胞生物学作用的影响。方法 采用体外细胞培养技术,通过 MTT测定法、3 H TdR 掺入法、成纤维细胞胶原凝胶模型研究 IFN- γ、TGFβ3对不同时期创面来源的成纤维细胞的增殖、DNA合成及其在胶原凝胶中收缩的变化。结果 IFN -γ和TGFβ3 在体外对正常皮肤真皮和烧伤后不同时期创面来源的成纤维细胞的生物学作用不同: (1)相同浓度的两种因子均能明显抑制正常真皮和伤后10、14 d创面成纤维细胞,对伤后28、35 d的创面成纤维细胞γ 干扰素的抑制作用较弱、TGFβ3 作用则不明显;(2)对成纤维细胞在胶原凝胶中的收缩能力,两种因子均有明显的抑制作用,而最强的作用均表现在对伤后21 d的创面成纤维细胞上。结论 γ -干扰素和转化生长因子β3 可能参与了调节创面成纤维细胞的表型转化与调控而影响其生物学特征,进一步的研究有利于为临床用生物学方法在创面愈合过程中控制瘢痕增生或挛缩提供参考依据。Objective To investigate the biological effectsof recombinant interferon-γ(IFN-γ) and transforming growth factor beta 3(TGFβ 3) on wound fibroblasts derived from different stages of human partial-thickness burn wounds. Methods Using the technique of cell culture, the probiferation, DNA synthesis and the contraction of fibroblast-populated collagen lattices (FPCL) were measured on the wound fibroblasts from different stages of burn wounds by means of MTT assay, 3H-TdR incubation and the vitro model of FPCL. Results IFN-γ and TGFβ 3 could significantly inhibit the proliferation and DNA synthesis on both normal skin dermal fibroblasts and wound fibroblasts of 10 and 14 days. But IFN-γ had little and TGFβ 3 had no effect on wound fibroblasts of 28 and 35 day. Both of two factors could inhibit the contraction of FPCL, and the most effective response was produced by wound fibroblasts of 21 days. Conclusion The different responses to IFN-γ and TGFβ 3 of wound fibroblasts might be due to the regulation of phenotypic properties by cell growth factors(cytokines). The work underway to estimate the further effects of cytokines on wound fibroblasts will be helpful to decrease hypertrophic scarring and/or scar contracture in human skin after deep partial-thickness burn injury.
