摘  要:目的研究急性粒细胞白血病患者白血病细胞内脱氧胞苷激酶 (DCK)的基因表达水平与阿糖胞苷 (Ara C)敏感性之间的关系。方法收集急性粒细胞白血病患者的骨髓标本 ,分离得到单个核细胞 ,采用RT PCR技术半定量测定DCKmRNA表达水平 ,比较敏感与复发耐药患者DCK表达水平的差异。结果复发耐药患者DCK的表达水平要明显低于敏感患者 (P <0 .0 1)。结论DCK表达的降低可能与急性粒细胞白血病患者对Ara C耐药相关 ,DCK表达水平可作为临床评价白血病患者对Ara C敏感性的一个指标 ,进而可以预测AraObjective To study the correlation of deoxycytidine kinase mRNA levels in acute myeloid leukemia (AML) and resistant leukemia patients. Methods By using semi-quantitative reverse transcriptase chain reaction (RT-PCR) technique, deoxycytidine kinase (DCK) gene expression in the bone marrow mononuclear cells of 30 patients was analyzed. The expression difference of DCK between sensitive and resistant patients was evaluated using statistical methods. Results DCK expression in the sensitive leukemia patients was much higher than that in the resistant one. Long time use of cytosine arabinoside (Ara-C) induced disappearance of DCK expression in some patients(P<0.01). Conclusion Low expression or deficiency of DCK represents one possible cause of resistance to Ara-C. Moreover, DCK expression may act as an indicator of Ara-C sensitivity and pharmacological effect.
