河南省HIV流行毒株env膜蛋白基因C_2-V_3区序列特征和亚型研究

摘　　要：目的　了解河南省内艾滋病病毒Ⅰ型 (HIV 1)流行株的亚型及序列变异特征。方法　用套式聚合酶链反应 (nested PCR) ,对河南省内 2 8份HIV 1感染者外周血单个核细胞 (PBMc)中前病毒脱氧核糖核酸 (DNA)的膜蛋白基因进行扩增 ,并对C2 V3 及其邻区 35 0～ 4 5 0个核苷酸序列进行测定和分析。结果　 2 8份样品间的基因离散率为 5 72 % ,与流行于云南省的 (泰国B)毒株基因距离为 5 80 % ,与欧美B亚型基因距离为 18 0 5 % ,而与其它A、C、D、F、G、H、J、O等亚型的基因距离均在 2 4 %以上。系统树分析显示 ,2 8份样本与泰国B亚型聚在一起而远离其它国际亚型。对于其V3 环四肽序列的分析表明 ,具有泰国B亚型基因型GPGQ的 8例 ,占 2 8 5 7% ;具有欧美B亚型基因型GPGR的 13例 ,占 4 6 4 %。结论　河南省流行的HIV株属B′亚型 ,其毒株来自与泰国接壤的云南省 ,在河南省流行的时间约在 6～ 10年 ,其V3 环顶端四肽序列特征以GPGR为主。objective To investigate the subtype distribution and sequence characteristics of HIV-1 strains prevalent in Henan province.Methods HIV-1 env gene was amplified by PCR from uncultured peripheral blood mononuclear cells(PBMCs)obtained from 28 HIV-1 seropositive individuals in Henan in 2002.The C 2-V 3 region(about 450bp)was sequenced.Results The innergroup gene divergence of 28 sample is 5.72%,and 5.80% with reference strains of Thailand B subtype from Yunnan,18.05% with North America B,over 24% with other subtypes including A,C,D,F,G,H,J,O.Phylogenetic tree analysis showed that 28 Henan B strains clustered closely to Thailand B and were far from other international subtypes.The analysis of V 3 loop amino sequence of 28 Henan B strains revealed that V 3-tip motif of 8 samples(28.57%)was GPGQ of Thailand B subtype gene,12 samples(42.86%)GPGR of Europe and America B subtype gene.Conclusion These findings suggest the epidemic of subtype B of HIV-1 in Henan province and it is closely related to those identified in Yunnan province.The data showed that these viruses existed for about 6-10 years.V 3 tip motifs of the HIV-1 isolates were mainly GPGR.
