摘  要:HPLC测定氨基酸,目前最为普及的衍生方法是邻苯二甲醛柱前衍生法,此方法最大的缺点是测不出具有亚胺基的氨基酸,如脯氨酸。 氯甲酸芴甲酯(9-芴甲氧羰酰Fmoc-Cl)是近年来用于多肽合成的新型氨基保护剂,它可以和全部氨基酸反应并生成荧光化合物,因此有可能做为分析全部氨基酸的荧光衍生剂。本文报告了以Fmoc-Cl对氨基酸做柱前衍生及反相色谱分离条件,并定量地测定了水解多肽中全部氨基酸的残基数。Fmoc (fluorenylmethyloxycarbonyl chloride) is a new aminoprotecting group, which has been applied in peptide synthesis. As Fmoc reacts with primary amine, as well as seconary amine, it can be used as a pre-column derivatizing reagent for analysis of total amino acids by HPLC We have investigated the chromatograrphic conditions of Fmoc-amino acids on reversed-phase HPLC and the optimal derivatizing procedure. The results have proved that the Fmoc protocol is a high sensitive, stable and quantitative method for analysis of amino acids in peptide hydrolysates. It is suitable for the analysts of amino acids, including some important ones which can not be detected in OPA procedure.
