吲哚美辛与沙立度胺联合治疗癌症恶病质的实验研究

摘　　要：目的　探讨联合前列腺素合成酶抑制剂 (吲哚美辛 )和免疫调节剂拮抗细胞因子 (沙立度胺 )治疗癌症恶病质小鼠是否优于单一应用。　方法　在接种 L A795肺腺癌细胞于 T739小鼠后 14 d建立癌症恶病质小鼠模型 ,将模型分成吲哚美辛 (Ind)、沙立度胺 (Tha)、(Ind+Tha)及生理盐水 (Sal) 4个组 ;腹腔注射给药 ,每日 1次 ,共 7d。另用正常小鼠作空白对照。于治疗前后采血分别检测细胞因子及营养指标水平。　结果　治疗前 Ind、Tha、(Ind+Tha)、Sal组的细胞因子水平均显著低于对照组 ,营养指标水平均显著高于对照组 (P<0 .0 1)。治疗后 Ind、Tha、(Ind+Tha)组细胞因子水平均显著低于 Sal(P<0 .0 5 ) ,营养指标水平均显著高于 Sal(P<0 .0 1) ;但 (Ind+Tha)的细胞因子水平均显著低于、营养指标水平均显著高于 Ind及 Tha(P<0 .0 5 )。　结论　通过拮抗细胞因子治疗癌症恶病质小鼠 ,吲哚美辛和沙立度胺联用比两药分别单用具有更佳的疗效。Objective To determine the better effect on cancer cachexia with a combination of indomethacine and thalidomide against cytokines as compared to any single one. Methods T739 mice inoculated with LA795 tumor cells were set as cancer cachexia models. The models were divided into four groups treated with indomethacine(Ind), thalidomide(Tha), indomethacine plus thalidomide(Ind+Tha), and saline(Sal) respectively, and normal mice were taken as control group(NOR). The serum levels of cytokine and nutritional marker were detected one day before(Pre 1) and seven days after(Post 8) the treatments. Results Before treatment, the levels of TNF-α and IL-6 in cancer cachexia models were significantly higher than those in NOR(P<0.01), and the levels of Glu, TC and Alb were significantly lower than those in NOR(P<0.01). The levels of TNF-α and IL-6 in Ind, Tha and (Ind+Tha) were significantly decreased than those in Sal, and the levels of Glu, TC and Alb in Ind, Tha and (Ind+Tha) were significantly lower than those in Sal(P<0.05, P<0.01). TNF-α and IL-6 in (Ind+Tha) were significantly lower than those in Ind or Tha, and the levels of Glu, TC and Alb in (Ind+Tha) were significantly higher than those in Ind or Tha(P<0.05). Conclusion That treatment of mouse cancer cachexia with a combination of indomethacine and thalidomide has a better effect than any single use.
