摘　　要：①目的　检测黑色素瘤抗原 1(MAGE 1)基因在胃癌组织中的表达 ,为应用MAGE 1基因产物对胃癌病人进行特异性抗肿瘤免疫治疗提供理论依据。②方法　用逆转录 聚合酶链反应 (RT PCR)检测 30例胃癌组织及相应癌旁组织中MAGE 1基因的表达 ,并对 1例RT PCR阳性扩增产物进行DNA序列测定。③结果　 30例胃癌组织标本中有 14例 (46 .7% )表达MAGE 1基因 ,而相应的癌旁组织均不表达MAGE 1,两组比较差异有显著性 (P =0 .0 0 1) ;DNA序列测定证实 1例RT PCR扩增产物中的目的基因片段为MAGE 1cDNA序列。④结论MAGE 1基因在胃癌组织中有较高表达 ,这使得以该基因编码蛋白作为疫苗用于胃癌的免疫治疗成为可能。Objective\ To investigate the expression of MAGE 1 gene in human gastric carcinomas and provide a rationale for the possibility of using MAGE 1 gene as a new peptide vaccine for immunotherapy of gastric carcinoma. \ Methods\ The expressions of MAGE 1 gene in 30 samples of human gastric carcinomas and 30 samples of corresponding normal tissue specimens were detected by reverse transcriptase polymerase chain reaction (RT PCR). One of MAGE 1 gene positive PCR products was sequenced.\ Results\ In the 30 gastric carcinomas studied, MAGE 1 mRNA was detected in 14 cases(46.7%). None of the corresponding normal tissue expressed MAGE 1 mRNA. The difference was significant( P =0.001). The DNA sequence confirmed that the target gene fragment was MAGE 1 cDNA. \ Conclusion\ MAGE 1 gene is expressed in a high percentage of gastric carcinomas. This tumor rejection antigen may provide tumor specific targets for immunotherapy of gastric carcinomas.
