摘 要:目的:建立大鼠血清中鬼臼毒素的高效液相色谱法(HPLC)测定方法。方法:用乙醚 -二氯甲烷(3∶1)萃取血清中鬼臼毒素,采用WatersSpherisorbODS2柱(5μm,4.6mm×250mm), 甲醇为流动相,流速1.0mL·min-1,二极管阵列检测器检测,波长为291nm。结果:血清中鬼臼毒素 含量在6.25~95.00μg·ml-1范围内呈良好线性关系,r=0.9995。方法回收率为90.4%~110.0%, 变异系数3%左右。结论:该方法测定血清中鬼臼毒素简便易行,准确可靠。Objective: To establish a HPLC method for the determination of the contents of podophyllotoxin in Serum. Methods: Detection was performed on Waters Spherisorb ODS2(5μm,4.6mm×250mm) by Photodiode Array Detector at 291nm with methanol(100%) as the mobile phase at a flow rate of 1.0mL/min, and podophyllotoxin of Serum was extracted with ether-dichloromethane(3:1). Results: The calibration showed a linear relation in the range from 6.25 to 95.00 μg·ml-1 with r =0.9995. the recovery rate of podophyllotoxin was 90.4%~110.0%. The relative standard deviation of podophyllotoxin was 3%. Conclusion: The method is an easy, accurate and reliable way to determine podophyllotoxin in Serum.
