Objective: To set up heroin addiction and readdiction model with the processes of heroin addiction→abstension treatment of heroin→heroin readdiction→reabstension treatment of heroin→heroin addiction a third time →abstension treatment of heroin a third time, and make clear the dynamic trend of the brain lesions. Methods:The changes of brain ultrastructures were observed under electron microscope and the changes of neurotransmitter such as NA, DA, 5-HT were analysed by means of fluorescence microspectro-photometry. Results: Degeneration, apoptosis and oncosis were found in the neuronal somas, axons, dendrites of the heroin addicted and readdicted rats' brains from all periods. Pathological changes of ultrastructures were found in the neuroglial cell and the neuropil. It showed the trend of aggravating by periodical pathological change. The changes of NA, DA,5-HT from the heroin addicted rats of all periods increased and it showed the trend of increasing with the increase of readdicting times. Conclusion: Widespread lesions have been found and were more severe with readdicted times. The neurotransmitters show pathological changes accordingly.
