摘　　要：目的探讨galectin 3mRNA在良性和恶性甲状腺肿瘤组织中的表达情况。方法对 8例甲状腺乳头状癌、4 5例良性腺瘤结节和 2 1例正常组织 ,采用Trizol抽提总RNA ,RT PCR扩增、凝胶电泳 ,检测galectin 3mRNA的表达 ,并进行统计学分析。结果galectin 3mRNA在甲状腺乳头状癌的表达为 10 0 % ,在良性腺瘤结节和正常组织的表达分别为 88.9%和 4 7.6 %。癌与腺瘤之间没有显著性差异 (P >0 .0 5 ) ,癌和腺瘤与正常组织之间均有显著性差异 (P <0 .0 5 )。结论galectin 3mRNA对于甲状腺癌的分子诊断不是一个适合的靶标 ,可作为甲状腺乳头状癌鉴定的辅助指标。Objective To study the relative expression levels of galectin-3 mRNA in benign and malignant thyroid tumor tissue as well as thyroid normal tissue. Methods The total RNA was extracted from eight papillary thyroid cancers (PTC), forty-five thyroid adenomas and nodes, twenty-one thyroid normal tissue. Complementary DNA was synthesized by polymerase chain reaction (PCR) and reverse-transcription (RT). The products were analysed by agarose gel and statistical approach. Results All PTCs had high level expression of galectin-3 mRNA .The expression of galectin-3 mRNA in thyroid adenomas and thyroid normal tissues was 88.9% and 47.6%, respectively. There was no evident difference between PTC and thyroid adenomas (P>0.05), but there was marked difference between PTC and thyroid normal tissues (P<0.05); it was also between PTC, thyroid adenomas and thyroid normal tissues (P<0.05). Conclusion Galectin-3 mRNA may not be a suitable molecular marker for thyroid carcinomas, although galectin-3 may be an accessory one in the diagnosis of papillary thyroid carcinomas.
