摘 要:目的探讨骨髓间充质干细胞(MSCs)移植促进大鼠脑缺血后神经功能恢复的可能机制。方法采用线栓法复制大鼠脑缺血2小时再灌注动物模型,实验动物分缺血对照组和移植治疗组,再灌注24小时后移植治疗组经颈动脉移植体外培养异体骨髓间充质干细胞。于移植后1周、2周、4周、12周分批处死实验动物,采用免疫组织化学染色及原位杂交技术观察缺血区脑组织的新生血管及VEGFmRNA表达的动态变化。结果①大鼠脑缺血后1周时缺血区皮层即可见大量的微血管增生,2周时达到高峰,4周、12周时有所下降。1周、2周、4周、12周时移植组比对照组缺血区皮层微血管密度(MVD)明显增高,P<0.05;②大鼠脑缺血后1周及2周时缺血区脑组织中均可见大量的VEGFmRNA阳性细胞,阳性信号主要分布于皮层及血管周围的神经细胞,缺血后4周及12周对照组VEGFmRNA阳性信号很弱,而移植组VEGFmRNA阳性信号仍较强。结论MSCs可能通过促进微血管增生改善缺血区血运修复缺血区损伤组织,从而改善脑缺血大鼠神经功能。Objective To probe into the mechanisms of bone marrow mesenchymal stem cells administration on neuro-functional recovery following focal cerebral ischemia in rats. Methods Forty healthy female rats were subjected to middle cerebral artery occlusion (MCAo) for 2 hours by intraluminal vascular occlusion, divided into MSC transplantation group and cerebral ischemia control groups at random. MSCs were administrated via carotid artery at 24 hours after MCAo in transplantation groups. At 1, 2, 4 and 12 weeks after MCAo, rats were killed in batches to investigate dynamically the regeneration of microvessels and VEGFmRNA expression in ischemic region by immunohistochemistry and situ hybridization. Results ①At 1 week after MCAo, a lot of microvessel proliferations in the injured cortex, reaching peak at 2 weeks, then decreased at 4 and 12 weeks after MCAo were observed. The MVD in the injured cortex of rats treated with MSCs was significantly higher than that of the control groups at 1, 2, 4 and 12 weeks after MCAo(P<0.05). ②Plenty of VEGF mRNA positive signals were found in cortical and perivascular neurons at 1 and 2 weeks after MCAo, and still showed more intensive in the MSCs transplanted groups than those in the controls at 4 and 12 weeks after MCAo. Conclusion The results suggest that MSCs transplantation can improve neurological function in cerebral ischemic rats through promoting revascularization in the injured region.
