摘 要:目的:探讨血管内皮细胞活性因子和系统性硬皮病(SSc)的关系及其在临床治疗中的意义。方法: 利用密度梯度方法检测循环内皮细胞记数(CEC),利用放射免疫分析检测血浆内皮素(ET)、血栓素B2 (TXB2)、6-酮-前列腺素F1α(6-K-PGF1α),利用Griss方法检测血浆一氧化氮(NO)水平,利用酶联免疫法 检测细胞间粘附分子-1(ICAM-1)、P选择素(P-S),本文对52例SSc病人作了治疗前后的血浆水平的检 测。结果:所有SSc患者均存在高CEC、ET、ICAM-1、P-S血症,存在低NO、6-K-PGF1α血症。结论:血管内 皮细胞活性因子的变化同SSc的发生发展密切相关,其血浆水平的检测有助于SSc发病机制的探讨,有助于 SSc疗效的观察。Objective To investigate the relationship between endothelial vasoactive factors and development of systemic sclerosis (SSc). Methods Blood circulating endothelial cell count (CEC, with density gradient precipitation method); plasma endothelin (ET), thromboxane B 2 (TXB 2), 6-keto-prostaglandin F 1α concentrations (all above three with RIA); nitric oxide (NO) contents (with Griss method) and plasma intercellular adhesion molecule-1 (ICAM-1), p-selectin (P-S) contents (with ELISA) were measured in 52 patients with systemic sclerosis (SSc) both before and after treatment with prostaglandin E 1 and 30 controls. Results Plasma ET、 TXB 2、 ICAM-1、 P-S contents and CEC were significantly higher and plasma NO, 6-K-PGF 1α contents were significantly lower in all the patients with SSc before treatment than those in the controls, After treatment, the patients with satisfactory responses (n=33) had the abnormal plasma values of these parameters greatly corrected (vs before treatment P<0.05, P<0.01); while poorly responding patients had the plasma values little changed. Conclusion Endothelial vasoactive factors are colosely involved in the development of systemic sclerosis and may be used as predictors of treatment efficiency.
