摘  要:目的 探讨Bcl 2反义寡核酸对膀胱癌细胞凋亡的影响。方法 以Bcl 2基因的前六个密码子互补的、以硫代磷酸修饰的寡核苷酸(PS ASO)作为实验组 ,以硫代磷酸修饰的Bcl 2正义寡核苷酸为阴性对照组 ,仅加入培养液的作为空白对照组 ,与人膀胱癌细胞株EJ细胞共同孵育 ,光镜、电镜观察细胞形态 ,流式细胞仪测定和分析。结果 与阴性对照组及空白对照组比较 ,Bcl 2PS ASO实验组细胞在第 1 2、2 4、48、72h出现较明显的凋亡峰 ,以及凋亡等形态学变化 ,凋亡率明显增高 (P <0 0 5) ,并显示时间的依赖性。结论 Bcl 2反义寡核酸能诱导膀胱癌EJ细胞凋亡 ,对膀胱癌的治疗有潜在的意义。Objective To investigate the effects of antisense oligonucleotides of BCL 2 gene on the apoptosis of bladder cancer EJ cell line.Methods The phosphorothioate antisense oligonucleotides (PS ASO) of BCL 2 gene (BCL 2PS ASO) with sequence targeted to the first six codes of BCL 2 gene as experiment group and sense oligonucleotides of BCL 2 gene (BCL 2PS SO) as negative control group and only culture as non control group were incubated with the EJ cell line. The apoptotic rate was assayed by flow cytometer and the cell form was observed with light and electron microscope. Results Compared with negative group and non control group, the experiment group presented transparent apoptotic cusp at 12, 24, 48, 72 h and occurred apoptosis under the microscope. The apoptotic rate was significantly increased in a time dependent manner ( P <0.05). Conclusions The PS ASO targeted the first six codes of BCL 2 gene could induce cells apoptosis, it might have potential significance in tumor therapy.
