摘 要:目的研究重度颅脑损伤大鼠采用脑源性神经营养因子(BDNF)和神经干细胞联合移植与神经干细胞单独移植治疗后神经功能的恢复及神经干细胞分化为神经元的比例,并探讨其可能的机制。方法将实验大鼠随机分为4组,Ⅰ、Ⅱ、Ⅲ组为BDNF和神经干细胞联合移植组(实验组),Ⅳ组为单独神经干细胞移植组(对照组),均制成大鼠重度颅脑损伤模型,分别行BDNF和神经干细胞联合移植或单独神经干细胞移植,对实验大鼠进行行为学观察,于伤后第2、5、8、11、14天抽取大鼠股静脉血测定一氧化氮(NO)含量,伤后2周处死大鼠后取脑组织行免疫组化染色观察。结果各实验组大鼠均较对照组神经功能恢复好(P<0.05或0.01),分化为神经元的比例较对照组高(P<0.05),NO的含量比对照组明显降低(P<0.01)。结论BDNF可能是通过降低损伤后NO的形成从而有利于移植的神经干细胞向神经元分化,BDNF和神经干细胞联合移植较单独神经干细胞移植对神经系统损伤有更好的治疗效果。Objective To evaluate the efficacy of brain-derived neurotrophic factor (BDNF) combined with neural stem cell transplantation in treatment of severe traumatic brain injury in rate. Methods The rats were randomly divided into 4 groups; 3 experimental groups and 1 control group. After the sever traumatic brain injury was induced, rats in experimental groups were treated with BDNF and neural stem cell transplantation, animals in control groups were treated with transplantation only. The behavior of animals was observed and the blood nitrous oxide (NO) levels were measured. After the animals were sacrificed the brains were examined immunohistochemistry. Results The recovery of nerve function in experiment groups was better than that in control group(P<0.05 or P<0.01); the differentiation ratio of neural stem cells to neuron in experiment groups was higher than that in control group(P<0.05)., the blood NO contents in experimental groups were lower than those in control group(P<0.01). Conclusion The combined treatment of BDNF with neural stem cell transplantation may have better therapeutic effect on head injury than the transplantation alone.
