青光眼术后5-Fu抗瘢痕形成的实验动物研究(兔眼房水的5-Fu药代动力学研究)

摘　　要：6只新西兰纯种白兔右眼行巩膜咬切术后,第三天球结膜下注射0.5%5～Fu 3mg。采用微量进样器对同一兔眼注射5-Fu前(0点)、注射后0.25、0.5、1、1.5、2、3、4、6、8、12小时不同时点多次采用作房水的5-Fu药代动力学研究。结果:注射后平均40分钟房水中5-Fu浓度吸收达高峰,高峰浓度平均为21.94μg/ml,半衰期平均为28.20分钟,4小时后5-Fu在房水中基本消除。Sclerectomy was performed on the, 6 right eyes of six New Zealand rabbits. On the third day after operation 0.5% 5-Fu (Fluorouracil) 3mg: was given subconjunctivaly. To test the pharmacokinetics of 5-Fu in the aqueous humor, a micro-sampler was used to aspirate the aqueous humor from the anterior chamber directly on each of the operated eyes prior and post to 5-Fu injection 0.25, 0.5, 1.0, 1.5, 2.0, 3.0, 4.0, 6.0, 8.0, and 12 hours, respectively. The results demonstrated that 21.94/ug/ml was the highest concentration of 5-Fu. in the aqueous humor at 40 minutes and the half-life time was 28.2 minutes postinjection. Four hours after subconjunctival injection 5-Fu totally disapeared.
