摘 要:目的观察大鼠下丘脑外侧区(lateral hypothalamic area,LHA)内注射五肽胃泌素(pentagastrin,G5)对大鼠胃缺血-再灌注损伤(gastric ischemia-reperfusion inimy,GI-RI)的影响。方法核团微量注射法,采用夹闭大鼠腹腔动脉30min,松开动脉夹血流复灌60min的GI-RI模型。结果①LHA内微量(0.5μl)注射G50.1、0.5、2.5ng后行胃缺血-再灌注(GI-R),其胃粘膜损伤面积分别较溶剂对照组增加41.3%、79.11%(P<0.01)和115.59%(P<0.01),且有剂量-效应依赖关系。②背侧迷走复合体(dorsal vagal complex,DVC)内微量(0.5μl)注射G52.5ng后GI-R,其胃粘膜损伤面积比DVC内注射等量溶剂后GI-R组增加69.15%(P<0.01)。结论LHA内注射G5对GI-RI具有加重作用,其作用机制可能是由于激活了LHA的胃泌素神经元,通过下行投射纤维作用于DVC神经元的胃泌素受体而起作用。Objective To investigate the effect of microinjection of pentagastrin (G5) into lateral hypothalamic area (LHA) on gastric ischemia-reperfusion injury (GI-RI) in rats. Methods Intranuclear microinjection and rat model of GI-RI were employed in the experiment. The model was made by clamping the celiac artery for 30 min followed by 60 min of reperfusion after removing the clamp. Results (1) Compared with the control group (with vehicle microinjection into LHA plus I-R), the extent of GI-RI in the test group after microinjection of G5 (0.1 ng/0.5 μl, 0.5 ng/0.5 μl and 2.5 ng/0.5 μl into the LHA) was increased by 41.3%, 79.11% and 115.59% respectively (P<0.01), displaying a dose-dependent manner. (2) After a microinjection of G5 (2.5 ng/0.5 μl) into the dorsal vagal complex (DVC), the GI-RI was increased by 69.15% (P<0.01) compared with the vehicle control. Conclusion The aggravative effect of microinjection of G5 into LHA on GI-RI might be mediated by the activation of gastrinergic neurons in the LHA, which release gastrin from the descending projection fibers and activate the gastrin receptors on the DVC neurons.
