摘 要:用鼻咽癌CNE-2Z细胞系行裸小鼠皮下移植,通过间接免疫酶标和间接免疫荧光方法,观察肿瘤移植后不同时间纤维粘连蛋白(fibronectin,FN)、层粘连蛋白(laminin,LN)和W型胶原(typeⅣcollagen,Ⅳc的分布,结果:1)FN的分布方式随肿瘤移植后时间的延长,从瘤巢间的散在点状或片状布分变为瘤巢间的纤维组织内和血管周围成纤维细胞增生区域的丝状或网状分布;2)LN和Ⅳ型胶原的分布方式基本一致,从肿瘤间质中血管壁和基底膜上的连续细线状分布变为不规则连续粗线状多层分布。而在肿瘤细胞侵袭部位的血管基底膜处则表现为LN和Ⅳ型胶原消失提示:在肿瘤发展过程中的不同时期,细胞外基质蛋白的分布存在差异,细胞外基质的分布及其合成和降解的变化在肿瘤的生长进程中具有一定的作用。Human Nasopharyngeal Carcinoma Cell Line CNE-2Z was implanted at thesubcutanous costal region of nude mice。 At different time intervals, local distribution ofnbronectin(FN),laminin(LN)and type Ⅳ collagen was detected by indirectimmunoperoxidase and immunofluorescence staining.It was shown :1) The characteristicpatterns of FN varied from the early punctual or fragmental pieces around nests of tumorcells to a later mesh-like network in the perivascular fibroblastic proliferation area and the fi-brous tissue around nests of tumor cells.(2) The distributing patterns of LN and type Ⅳcollagen were virtually identical. Their distribution varied from the continuously thin line onlyinside the walls of all types of blood vessels in the tumor matrix to the irregular and continu-ous thick line spreading over the whole vascular and perivascular area, whereas at the invasivelocus of basement membrane such components disappeared with progression of invasion。The results mentioned above indicate that the distribution of extracellular matrix differsat different times during tumor progression。It shows that the distribution of extracellular ma-trix and the changes in synthesis and degradation of the extracellular matrix components mayplay an important role during malignant tumor progressive growth。
