制滴菌素A治疗人肺腺癌细胞A549裸鼠移植瘤的实验研究

摘　　要：目的　探讨组蛋白去乙酰化酶抑制剂制滴菌素A (trichostatinA ,TSA)对人肺腺癌细胞A5 4 9裸鼠移植瘤的治疗作用。方法　建立人肺腺癌细胞A5 4 9裸鼠模型 ,瘤内注射TSA和 0 9%生理盐水 (NS) ,对治疗前后的裸鼠进行瘤重量、瘤体积、细胞形态和凋亡的观察。结果　在实验组和对照组之间 ,瘤重量差异有显著性 (P <0 0 5 ) ;TSA治疗 1周和 2周后瘤体积与治疗前瘤体积相比 ,差异无显著性 (P >0 0 5 )。NS治疗 1周后瘤体积与治疗前相比差异无显著性 ,而治疗 2周后瘤体积与治疗前相比 ,差异有显著性 (P <0 0 5 )。电镜下可见肿瘤细胞的凋亡现象经TSA干预后明显增多。结论　TSA治疗人肺腺癌细胞A5 4 9裸鼠模型有效。Objective To explore the therapeutic effectiveness of trichostatin A(TSA ) in the treatment of transplantable mode of human lung adenocarcionma A549 cells in nude mice. Methods Nude mice mode bearing lung carcinoma was established using human adenocararioma A549 cells. TSA or 0 9 % normal saline (NS) were injected into the tumor. The weight of neoplasia, the volume of neoplasia, and the form of cells were observed under light and electron microscopes. Results The difference of the average weight and average volume were statistically significant between TSA group and NS group at the end of experiment ( P <0 05); Before and one week after experiment, the average volume of the tumor tissue showed no statistically significant difference in NS group, P >0 05; but the difference were statistically significant between one week and two week treatment in NS group. The difference of the average volume and weight had no statistically significant difference in TSA group before and after treatment, P >0 05. The apoptosis of the tumor cells were found more under electron microscopy in TSA group than in NS group. Conclusion TSA is effective in the treatment of adenocarcinoma in nude mice.
