苯妥英钠对慢性应激大鼠大脑前皮质诱导型一氧化氮合酶表达的影响

摘　　要：目的:探讨慢性应激对大鼠大脑前皮质诱导型一氧化氮合酶(induciblenitricoxidesynthase,iNOS)表达的影响及苯妥英钠于其中的效应,分析与情感障碍发生的关系。方法:实验于2000-05/08在汕头大学医学院精神病学实验室进行。取32只SD大鼠,随机分为对照组、应激组和应激给药组。采用强迫游泳作为应激模型,利用免疫组织化学方法及计算机图像分析技术,定量测定各组大鼠大脑前皮质iNOS的表达水平。结果:应激给药组大鼠大脑前皮质iNOS表达的平均灰度值(161.76±3.01)显著高于应激组(156.99±2.18),而与对照组(158.33±5.95)比较差异无显著性意义;应激组大鼠大脑前皮质iN-OS阳性表达的相对切面面积比犤(5.45±2.18)%犦显著高于对照组犤(0.62±0.47)%犦和应激给药组犤(0.58±0.43)%犦,后两组间差异无显著性意义。结论:苯妥英钠能阻止慢性应激所致的大鼠大脑前皮质iNOS表达水平增加。AIM:To investigate the changes of inducible nitric oxide synthase(iNOS) expres sion levels in rat prefrontal cortex induced by chronic stress, and analyze the relation between the effects of diphention(DPH) on them and feeling disorder. METHODS:The study was processed in the Department of Psychiatry, Shantou Unive rsity Medical College from May 2000 to August 2000.Thirty two adults male SD ra ts were randomly divided into three groups:control group,stress group and stress DPH group.The forced swimming was performed to stress rats.Using the immunohi stochemistry and the computerized image technique,the expression levels of iNOS in rat prefrontal cortex were assayed quantitatively in each group. RESULTS:The iNOS average grey degree in the prefrontal cortex of stress DPH g roup(161.76±3.01)was significantly higher than that of stress group(156.99± 2.18),and had no significant difference as compared with control group(158.33 ±5.95).The iNOS relative sectional area proportion in prefrontal cortex of stre ss group[(5.45±2.18)%]was significantly larger than that of control group[ (0. 62±0.47)%]and stress DPH group[(0.58±0.43)%].There was no significant dif ference in iNOS relative sectional area proportion between control group and str ess DPH group. CONCLUSION:Diphentoin can prevent the enhancement of iNOS expression in prefro ntal cortex of rates induced by chronic stress.
