摘  要:目的:探讨预先针刺是否能诱导脑缺血耐受而具有脑保护作用及对神经元凋亡的影响。方法:实验在黑龙江中医药大学神经解剖教研室进行。取40只Wistar大鼠随机分为正常对照组、假手术组、脑缺血组、针刺预处理组各10只。针刺预处理组脑缺血前给予针刺,采用“4-动脉阻断”方法建立大鼠全脑缺血模型。神经元内氏体亚甲兰特殊染色法观察大鼠脑缺血后海马CA1区神经元内氏体变化;原位细胞凋亡检测法TUNEL染色)及(电镜观察脑缺血后大脑皮质、海马CA1区神经元凋亡情况。结果:脑缺血组皮质、海马CA1区可见大量棕色TUNEL染色阳性细胞核,皮质为(65.36±10.18)个/视野,海马CA1区(58.34±9.64)个/视野,针刺预处理组阳性细胞核皮质(10.63±5.39)个/视野,海马CA1区为(12.16±5.69)个/视野,较脑缺血组明显减少(q=6.55,8.73,P<0.05),内氏体染色阳性细胞数脑缺血组皮质为(16.16±8.31)个/视野,海马CA1区(21.66±9.39)个/视野,针刺预处理组阳性细胞核皮质(55.36±12.19)个/视野,海马CA1区为(79.36±10.69)个/视野,较脑缺血组明显增多(q=7.65,9.23,P<0.05)。结论:针刺预处理可以诱导脑缺血耐受,减轻缺血后神经元损伤,抑制神经元凋亡可能是预先针刺发挥脑保护作用的一种途径。AIM:To study whether pre acupuncture can induce brain ischemic tolerance to p rotect brain and its influence on apoptosis. METHODS:The experiment was done in the Department of Anatomy,Heilongjiang Univ ersity of Traditional Chinese Medicine.Forty Wistar rats were randomized into co ntrol group,sham operated group,cerebral ischemia group and pre acupuncture gr oup.Acupuncture was performed in the rats of acupuncture group before cerebral i schemia,and four vessel occlusion(4 V0) was used to establish rat model of glo bal cerebral ischemia.The neuronal Nissl staining was used to observe the change s of Nissl body in hippocampal CA1 region,TUNEL staining and electron microscope used to observe the apoptosis of cerebral cortex and hippocampus after cerebral ischemia. RESULTS:By TUNEL method, many brown positive nuclei were seen which were(65.36 ±10.18) per visual field in cerebral cortex and(58.34±9.64) per visual field i n hippocampal CA1 region of the ischemic group,and(10.63±5.39) per visual field and(12.16±5.69) per visual field in the pre acupuncture group respectively,si gnificantly decreased as compared with the ischemia group(q=6.55,8.73,P< 0.05).B y neuronal Nissl staining, positive neuron were seen which were(16.16±8.31) per visual field in cerebral cortex,(21.66±9.39) per visual field in hippocampal C A1 region of the ischemic group,and(55.36±12.19) and(79.36±10.69) per visual f ield in the pre acupuncture group respectively,significantly increased as compa red with the ischemia group(q=7.65,9.23, P< 0.05). CONCLUSION:Pre acupuncture can induce brain ischemic tolerance so as to relie ve post ischemia injury, and inhibiting apoptosis may be a way of pre acupunct ure to protect brain.
