摘 要:目的 观察人胃癌细胞系SGC7901瘦素及瘦素受体的表达及瘦素对SGC7901增殖及凋亡的影响。方 法 免疫组化和RT-PCR检测SGC7901瘦素和瘦素受体的表达,噻唑蓝比色实验观察瘦素对SGC7901的增殖 作用,流式细胞仪检测瘦素对细胞周期及凋亡的影响,RT-PCR方法半定量检测瘦素对促凋亡基因bax mRNA 表达的影响。结果SGC7901既表达瘦素又表达瘦素受体,重组瘦素呈剂量依赖性地促进细胞的增殖,并明显 抑制细胞的凋亡,瘦素可抑制促凋亡基因bax mRNA的表达。结论 瘦素可能以自分泌方式调节人胃癌细胞系 SGC7901的增殖及凋亡。Purpose To detect the existence of leptin and leptin receptor in human gastric cancer cell line SGC7901 and to investigate the effect of leptin on proliferation and apoptosis in SGC7901 cell line. Methods The expression of leptin and leptin receptor was detected with the immunohistochemistry and RT-PCR respectively. The proliferative effect of leptin was investigated with MTT assay and the apoptosis of cell was measured with flow cytometry. The effect of leptin on the expression of bax mRNA was measured with RT-PCR in semi-quantity. Results Leptin and leptin receptor was expressed in SGC7901 cell line. Leptin could promote the proliferation and inhibit the apoptosis of SGC7901 cell line in a dose-dependent manner. Leptin could inhibited the expression of bax mRNA. Conclusions Leptin may play an important role in human gastric cancer by regulating cell growth and apoptosis.
