原癌基因c-erbB-2扩增、过表达与肾癌生物学行为的关系

摘　　要：目的 :探讨原癌基因 c- erb B- 2扩增与过表达在肾腺癌的临床意义。方法 :应用原位杂交和免疫组化对 90例肾癌组织 c- erb B- 2扩增与过表达进行研究。结果 :在 90例肾癌中 ,8例显示 c- erb B- 2基因扩增 ,2 4例显示 c-erb B- 2蛋白过表达。基因扩增与蛋白过表达呈显著相关关系 (P<0 .0 5 )。扩增与蛋白过表达均与肾癌病理分级、临床分期及淋巴结转移呈正相关。结论 :c- erb B- 2基因扩增和过表达均可成为判定肾癌预后的指标。c- erb B-Objective:To investigate the amplification and overexpression of c-erbB-2 oncogene in renal cell carcinoma.Method:Renal cell carcinoma spe-cimens from 90 patients were studied by in situ hybridization and immunohistochemistry for c-erbB-2 amplification and overexpression.Result:In 90 renal cell carcinoma,8 specimens were found to have amplificated c-erbB-2 gene,where as 24 specimens were found to have overexpression of the c-erbB-2.Amplification of c-erbB-2 gene was of positive correlation with overexpression of the gene( P <0.05).[WT5”,6BZ]It was found that the amplification and overexpression of c-erbB-2 oncogene was of positive correlation with the grades,stages,and degree of lymphatic metastasis of renal cell carcinoma( P <0.05).Conclusion:Both the amplification and the overexpression of the protooncogene c-erbB-2 might be significant indicators for evaluating the prognosis of renal cell carcinoma.The overexpression of c-erbB-2 might provide more clues than the amplification of genes.
