摘 要:大麦黄花叶病是由禾谷多粘菌(polymyxa graminis)传布的一类麦类病毒病。是上海市效区及长江中下游及沿海大麦产区的重要病害。大麦黄花叶病毒(BaYMV)在国内外虽已有不少研究,但对国内分离株的生化性质尚未有详细报道。本文报道以上海郊区的BaYMV为研究材料,成功地建立了一套分离提纯的方法,并在此基础上,研究了该病毒的结构蛋白和核酸性质,并和国外的一些研究结果进行了比较,从而为今后应用生物技术防治大麦黄花叶病毒打下了基础。The Barley Yellow Mosaic disease is an important crop disease spread in the suburbs of Shanghai and coastal arears in East China. A purification procedure of Shanghai isolate of BaYMV was developed using the combination method of organic solvent treatment,ultracentrifugation,sucrose and CsCl densi- ty gradient centrifugation. The isolate showed the existence of two peaks in particle-length distribution (300 nm and 600 nm). SDS-PAGE of purified virus preparation demonstrated one major band of 29kd. A minor band of 33kd appeared occasionaly. The density of 33kd band could be increased signifi- cantly,when two protease inhibitor PMSF or laupeptin were added druing extraction and purification. Besides two major species of RNAs of the isolate with size about 2.78×10~6 and 1.12×10~6,one small RNA about 0.68×10~6 were observed in non-denaturing agrose electrophoresis. The relationship be tween the Shanghai isolate and other isolates of BaYMV reported previously was discussed.
