摘  要:选用人胚肺为实验材料,研究了十字花科蔬菜提取物oltipraz对人肺癌病变早期细胞恶性转化的阻断作用。将阻断按时间分为oltipraz在CSC作用前、后和同时三组,并设CSC、oltipraz、DMSO三组对照,将上述6组体外培养处理的人胚肺组织,提取DNA转染Rat-1细胞,用低血清筛选、作软琼脂培养、裸鼠接种鉴定,结果在CSC、CSC→oltipraz两组获得恶性转化细胞,CSC组恶性度更大。提示oltipraz对CSC所致的细胞恶性转化具有一定阻断作用,oltipraz在CSC运用前或同时运用效果较好;进一步推断oltipraz对人肺癌变早期具有一定防治作用。Human fetal lung (HFL) as experimental material was selected and inhibition of celluar malignant transformation on HFL carcinogensis at early stage by oltipraz,dithiolthine present in cruciferous vegetables,was studied.Inhibition was divided imto three groups:oltipraz prior to CSC,oltipraz at same time CSC,oltipraz after CSC.To contrast,negative control groups CSC,oltipraz,DMSO were set up.These six groups cultured tissue was extracted for DNA and transfected into Rat 1 cell.Then it was operated by low serum selection;through soft agar culture,nude mice carrying tumor,malignant of cell was identified.Resuluts show that CSC and CSC→oltipraz groups gain transfected Rat 1 cell (T rat 1),T Rat 1 cell of CSC groups is more malignant.These results support the effective inhibition of early HFL carcinogensis on celluar malignant transformation and it is effective of oltipraz prior to or at same time CSC treatment.It is concluded that oltipraz can prevent and treat early human lung carcinogenesis.
