摘 要:本文以简化的方法从伤寒沙门氏菌Ty2菌苗株制备外膜蛋白(Outer membraneprotein,简写为OMP),OMP的生化检测显示主要为蛋白质,并有少量LPS污染,不含有Vi多糖。由SDS-PAGE显示OMP为一组分子量介于17~70KD的蛋白质,其中分子量为36~41KD左右的蛋白带为主要蛋白带。OMP有良好的免疫原性和抗原性,不加佐剂免疫动物可产生高效价抗血清,玻片凝集试验能与不同类型抗原结构的伤寒沙门氏菌凝集,但这种凝集可因细菌Vi抗原的存在而被阻抑。动物试验表明OMP有良好的免疫原性,自动和被动免疫力试验均能对小鼠有良好的保护作用,表明OMP是一种保护性抗原。动物热原试验和体重减轻试验及毒性试验均符合有关要求。This paper described a simplified method for preparing the outer membrane proteins (OMP) with the strains Ty2 of S. typhi. Biochemical analysis showed that the main content of the OMP was protein with minor contamination of LPS, but no Vi polysaceharide was found. SDS-PAGE Profile showed that the OMP was composed of a number of different protein bands with molecular weights ranging from 17 to 70 KD, but the molecudlar weights of most of proteins were 36~41KD. The OMP showed good antigenicity and immunogenicity. Animals immunized by the OMP without adjuvant could induce high titre anti-OMP in serum. Agglutination test on glass slide showed that S. typhi of various types of antigen construction could be agglutinated with anti-OMP serum, but the agglutination could be inhibited by Vi antigen. Both active and passive immunity tests demonstrated good protection of the OMP to mice. Thus, it suggested that OMP was a protective antigen. Additional animal tests were done in regard to the toxicity, body-weight decreasing effect as well as pyrogenicity and all results could meet the conventional requirements. The OMP prepared as described above might be used as a candidate for the development of a new generation of typhoid vaccine.
