摘  要:以4个水稻品种的成熟胚诱导的愈伤组织为材料,研究了继代培养中胚性与非胚性愈伤组织的游离氨基酸库的变化和愈伤组织再分化的关系,结果表明,天冬酰胺含量随愈伤组织继代培养时间的延长而成倍增加,而酪氨酸、鸟氨酸和氨含量则显著减少,品种之间表现一致,与愈伤组织分化率的相关系数达显著或极显著水平,胚性愈伤组织经较长时间的继代培养后与未继代培养的非胚性愈伤组织有相似的游离氨基酸库,同时表明继代培养过程中愈伤组织游离氨基酸库中酪氨酸、鸟氨酸和天冬氨酸的变化导致胚性丧失,可通过添加外源氨基酸加以调控。In this paper, correlation of changes in the free amino acid pool withredifferentiation for mature embryo callus subculture in Oryza sativa L. were reported.Along with the time of subculture are increased, asparaging content of calli increased,but the contents of tyrosine, ornithine and NH_3 of calli decreased. The correlationcoefficients between callus differentiation frequency and content of tyrosine, orornithine, or NH_3 or asparaging were significant at 5% or 1% probability level,respectively. The free amino acid pool of long-term subcultured embryogenic callus issimilar as that of non-embryogenic callus. It were showed that the growth and devel-opment of embryogenic callus could be regulated via subculture medium supplementedwith various amino acids.
