摘 要:本实验研究了二乙基二硫代氨基甲酸钠(DDC)一血卟啉衍生物(HPD)—激光及DDC—放射线的抗癌作用。其结果表明,DDc可显著地提高HPD—激光的抗癌作用,使癌细胞死亡率从15.9%提高到92.9%(P<0.01)。对荷瘤小鼠,先给DDC也可提高其疗效。用ESR法,自旋捕捉剂与自由基清除剂研究的结果证明,DDC提高HPD—激光的抗癌作用与增强癌细胞内自由基反应有关。DDC亦可提高γ射线的癌细胞(S180)的杀伤作用,也能提高电子直线加速对小鼠(BALB/C)EMT6/Ku实体瘤的疗效。We have studied the antitumor effects of DDC-HPD-laser and DDC-ionizing radition. In invitro studies, the death rate of tumor cells(H22)teated with DDC-HPD-laser was 92.2%.Whereas that of the cells treated with HPD-laser was on only 15.9%. These results show that DDC can significantly enhance the tumor cell-killing effect of HPD-laser (P<0.01 ).In invivo studies,the preliminary result indicates that the treatment with DCC-HPD-laser may be beneficial to tumor-bearing animals. The free radicals in tumor cell suspensions were trapped by DMPO and detected by ESR spectra.The role of free redicals was observed with free radical scavengers. The results of these studies shoW that the enhancement of antitumor effects of HPD-laser by DDC resulted from the enhancement of free radical reactions in tumor cells. Instead of HPD -laser we also used ionizing radiation to generate oxyge-derived radicals and found that DDC also enhanced the killing effect of γ-ray upon tumor cell(S180) and the antitumor effect of ionizing radiation produced by linear accelerator upon transplant EMT6/ku tumor on BALB/cmice.
