胃癌和结肠癌中rasP^（21）表达的研究

摘　　要：本研究以ｒａｓ癌基因表达产物Ｐ２１的单克隆抗体，利用ＡＢＣ免疫组化方法检测胃癌、结肠癌中Ｐ２１的表达情况。结果显示：检测的３８例胃良性病变中，除４例慢性萎缩性胃炎呈阳性反应外，其余病例全部为阴性；３４例胃癌全部为Ｐ２１表达阳性；１８例良性结肠病例中，除２例反应呈弱阳性外，其余为阴性；２８例结肠癌除外一例反应呈阴性反应外，全部为Ｐ２１表达阳性。此结果提示：在胃癌、结肠癌组织中存在着特异性的Ｐ２１表达；ｒａｓｐ２１可作为诊断胃癌、结肠癌的一个较为可靠的标志物、用ＡＢＣ免疫组化方法检测ｒａｓ癌基因产物可能为临床上诊断胃癌、结肠癌提供一种新手段。This study was undertaken to determinhe the expression of ras on-cogene product p2l in gastric and colonic arcinoma,using ABC immunohistochemicalassay with p21 monoclonal antibody. The result showed that benign gastric tissue obt-ained from normal gastric mucosa or benign gastric lesions reacted negatively in 42 outof 46 cases except four cases of chronic atrophic gastritis; all 34 cases of gastri ca-rcinoma stained positively for p21, Similarly,benign colonic tissue from normal colonor benign colonic lesions were negative for p2l in 21 out of 23 cases,colonic carinomastained positively for p2l in 27 out of 28 cases.The results recorded in this study suggestedthat there was expression especially of ras p2l in gastric and colonic carcinoma. Rasp2l might be used as a tumour marker of gastric and colonic carcinoma. The immuno-histochemical assay for the oncogene product may prove o be a new method for thehistodiagnosis of gastric and colonic caruinoma.
