摘 要:将穆斯堡尔(Mossbauer)γ射线共振吸收技术用于细胞灭活,称为穆斯堡尔灭活法。它具有剂量低、共振强的优点,避免了放疗中大剂量照射引起的严重的副作用。作者引入正铁血红素(Hematin)作为铁原子的载体,用穆斯堡尔灭活法,对白血病自身骨髓移植中的骨髓体外净化问题进行研究。通过在样品中加入不同浓度的Hematin,观察到Hematin对正常骨髓细胞、CML骨髓细胞和K562细胞的不同作用,测量了Hematin和γ射线共同作用下的细胞灭活效率。本文详细地介绍了穆斯堡尔灭活原理、实验方法和实验结果。The low-dose gamma-ray resonant Mossbauer radiation was used for the neoplastic cell inactivation, called Mossbauer inactivation. It has an advantage over the conventional radiotherapy in reducing hazardous side effects caused by high dose radiation, and it would be a new purging approach for autologous bone marrow transplantation (ABMT).In the experimental research, hematin was selected as the carrier of the 57Fe(the Mossbauer target). Bone marrow cells collected from normal donors and patients with chronic myelogenous leukemia (CML) and leukemic cell line (K562) have been investigated after the treatment with different concentration of hematin. The effect on the growth inhibition of clonogenic cells was observed in bone marrow cultures and leukemic cell line(K562), with the combined effect of Hematin and γ radiotion. The details of the principle of Mossbauer inactivation, experimental method, and experimental resrlts are given.
