胃癌p^（53）突变产物的增强表达

摘　　要：作者应用ＡＢＣ免疫组化方法，对３８例手术切除的胃癌组织及癌旁胃粘膜的冰冻切片标本进行了ｐ５３突变蛋白表达的检测．结果：３８例胃癌标本，２４例ｐ５３突变蛋白高表达，阳性率６３．２％，高分化腺癌３／４（７５．０％）阳性，中分化腺癌１３／２０（６５．０％）阳性，低分化腺癌５／９（５５．６％），粘液腺癌３／５（６０．０％）阳性，癌旁胃粘膜有１０例ｐ５３突变蛋白弱表达．２３例伴有淋巴结转移的胃癌标本，１８例（７８．３％）ｐ５３蛋白高表达．以上结果提示：抑癌基因ｐ５３的突变在胃癌的发生中是一个比较常见的基因改变，尤其在分化型腺癌的发生中表现明显．By using ABC immunohistochemical method,we studied expression of p53 protein in cryostatic sec-tions of tumor tissues and their adjacent mucosa resected from 38 patients with gastric cancer.p53 was found positive in nuclei with intensive staining in 24 out of 38 cases with carcinoma(63.2%),in which 3/4 were well differentiated,13/20 moderatelly differentiated,5/9 poorly differentiated and 3/5 mucinous adenocarci-noma. p53 positive cells were distributed diffusively in the tumor tissues. All the adjacent mucosa except 10 were negatively stained with anti-p53 monoclonal antibody pAb1801.These 10 adjacent mucosas were only weakly stained.p53 was also found positive in 18 out of 23 cancer patients with metastasis in perigastric lymph nodes(78.3%).The results suggest that p53 inactivation may be one of the genetic events in development of gastric carcinoma and may play a role in tumor metastasis.
