摘　　要：本研究通过对比不同凝胶组成成分及电泳条件，建立了非同位素ＰＣＲ－ＳＳＣＰ方法以检测ｐ５３基因第４外显子第７２密码子的多态性。对３５例正常组织ＤＮＡ的研究结果显示，ｐ５３基因这位点的杂合率和纯合率分别为４８．６％（ＣＣＣ／ＣＧＣ），４５．７％（ＣＧＣ／ＣＧＣ）和５．７％（ＣＣＣ／ＣＣＣ）．本法简单、快速，能有效地检测基因多态性，并可进一步应用于对肿瘤细胞ｐ５３基因杂合性缺失的分析。By comparing the different gel compositions and electrophoresis conditions ,the non-isotopic PCR-SS-CP method was developed for detecting the polymorphism in exon 4 of p53 gene. The results showed thatthe three genetic structures of the DNA fragment containing the polymorphic site could be clearly distin-guished by thismethod. The frequency of heterozygosity and homozygosity in 35 cases were 48.6%,45.7%(CGC/CGC)and 5.7%(CCC/CCC) respectively. Our results indicate that the non-isotopic PCR-SSCP is a simple ,fast and effective method for the analysis of polymorphism. It can be used for furtherstudy of loss of heterozygosity of p53 in human tumours.
