摘  要:以2%的蝌蚪提取液培养小鼠红白血病细胞(MELC)。结果:生长曲线显示蝌蚪提取液可抑制MELC生长;联苯胺染色反应显示MELC胞浆中血红蛋白的合成随提取液作用时间的延长而增加,说明蝌蚪提取液有诱导MELC发生终未分化的作用;流式细胞仪分析证明蝌蚪提取液可造成G0和G1期细胞堆积,G2和M期细胞减少。另经SDS-聚丙烯酰胺凝胶电泳分析:该提取液中的有效作用物质是一组分子量7万Da左右的肽类,在本提取液中的含量为4.5g/L。结果提示:蝌蚪提取液起诱导作用的成分易纯化,可能成为研究肿瘤恶性逆转的良好诱导分化剂。The murine ervthroleukemia cells(MELCs) cultured with the extract[1:50(v/v)concentration]from tadpoles(T-871) were studied in this experiment.The results showed that T-871 could inhibit the growth of the MELCs and the reaction stained with benzidine hydrochloride showed that the amount of hemoglobin synthesized was increased as the exposure time lengthened.T-871 could induce MELCs terminal differentiation.The results analyzed with flow cvtometry(FACS-420)demonstrated that the number of cells accumulated at G0+G1 phases and the number of cells decreased at G2 +M phases.The SDS-PAGE showed that a group of molecular weights of T-871 in effect on MELJCs were about 70 000 dalton and its chemical nature was of peptide.The effective contents of T-871 were 4.5 g/L. In our study,it was suggested that the effective components induced by T-871 be easily purified and could become good inducer for research on malignant cell reversion.
