摘  要:本文观察了低氧对离体培养成年牛肺动脉平滑肌细胞(PASMC)3H-TdR参入的影响,并用异搏定、H7、PMA和ECF进行干预实验,对SMC增殖中信息传递途径作了初探。结果显示,(1)缺氧24h,PASMC3H-TdR参入显著增高。(2)异搏定及H7显著降低低氧H-SMC3H-TdR参入,而PMA及ECF显著增加HSMC3HTdR参入。(3)H-SMC对PMA和ECF反应强于常氧N-SMC;但对异搏定及H7反应弱于N-SMC。结果提示:低氧可直接刺激成年牛PASMC增殖,其途径可能包含Ca2+内流增加和PKC进一步活化。外源PMA可能会进一步激活PKC而增强SMC增殖。EGF促增殖作用与PKC有一定关联。NSMC及H-SMC对干预物质反应程度的差别,反映出低氧是多环节作用于细胞代谢,从而提高细胞整体代谢水平。he effect of hypoxia on incorporation of 3H-TdR into PASMC of bovine invitro was observed.Verapamil,H7,PMA ,and EGF were used for interfering ex-periments to study signal transduction pathway in PASMC prolifetation.Theresults showed that(1)after 24h of hypoxic exposure,the incorporation apparentlyincreased;(2) in hypoxic PASMC , verapaxmil and H7 could notably decrease theincorporation and PMA and EGF could notably increase the incorporation;(3)the reactions of hypoxic PASMC to PMA and EGF was more intense than that ofnormoxic PASMC, but its reactions to verapamil and H7 was weaker than that ofnormoxic PASMC.The results indicated that PASMC could proliferate under hypoxic stmulation by means of increasing Ca2+ influx and further activating PKC;foreign PMA might enhance the hypoxic proliferation by activating PKC;the hypo-xic proliferation promoted by EGF might have some relation with PKC pathway; inconsideration of the different reactive levels to signal transduction interferingbetween normoxic and hypoxic PASMCs, it is possible that hypoxia might increasethe whole metabolism of PASMC through acting many metabolic links.
