有色金属混合尘诱癌病变图象分析及DNA含量测定的研究

摘  要:采用图象分析仪对有色金属混合尘诱发肺腺癌过程中正常、增生支气管上皮、癌前病变和肺腺癌(1~4组)进行细胞形态计量及DNA含量测定,结果显示:经逐步判别分析,从16项几何参数中优选出对各组诊断、鉴别诊断有较大价值的7项参数,并建立判别函数方程,经回代判别分析,准确率达96.3%;DNA含量测定,1,2组未出现DNA倍性>5C细胞,3组DNA倍性>5C细胞为1.25%,4组为5.0%。两组间存在极显著差异(P<0.01).表明形态计量及DNA含量测定对实验性肺腺癌研究有较大的实用价值。A computer-assisted image analysis system was applied to quantitate the cellular morphometry and DNA content of normal epithelia of bronchi (Group 1),hyperplastic epithelia (Group 2),atypical hyperplastic epithelia (Group 3) and adenocarcinomas (Group 4)in the course of the experimental lung adenocarcinoma induced by non-ferrous metal dust.Among 16 geometrial parameters,7 were chosen for diagnosis and differential diagnosis of lesions of the four groups.Each value was tested by stepwise discrimination classification. the accuracy rate being 96.3%. DNA ploid was investigated, and no case of >5 cell was found in normal and hyperplasis. Severe atypia had a lower percentage (1.25 %) and Lung adenocarcinoma was characterized by a high perecentage of > 5 cell (5.0%). The difference in percentage of > 5 cell between the severe atypia and adenocarcinoma was statistically significant(P<0.01).It is concluded that the cellular morphometry and DNA content measurement is very helpful to the pathological diagnosis of experimental lung adenocarcinoma.
