单克隆抗体体外大容量骨髓净化白血病的研究

摘　　要：应用补体介导细胞毒法体外消除２例白血病骨髓中残存瘤细胞，并作自身骨髓移植。用国产ＣＤ７单抗（ＬＮ６７－８及ＬＳ９３－２，ＩｇＧ２ｂ）首次净化１例Ｔ－ＡＬＬ骨髓及ＣＤ１０单抗（５５，ＩｇＧ２ａ）净化１例ＣＡＬＬＡ＋－ＡＬＬ。靶细胞清除率均达３个对数级，骨髓ＣＦＵ－ＧＭ回收率分别为５７．６％～７５．８％及５１．７％～７２．３％。Ｔ－ＡＬＬ病人移植后２６天获造血重建。现已完全缓解３年，首次证实国产单抗ＬＮ６７－８及ＬＳ９３－２净化骨髓有效、安全。Two patients with leukemia were autografted while purging of residual leukemic cells of bone marrow with complement mediated cytotoxicity. One patient of T-ALL was purged with CD7 monoclonal antibody (LN67-8 ,LS93-2, IgG2b),the other of CALLA+-ALL with CD10 monodonal antibody (55, IgG2a.). The Purging efficaly was as high as three common logarithmically grade pecovery rate of bone marrow CFU-GM was 57.6%￣75.8% and 51.7%￣72.3% respectively. The patient with. T-ALL reconstituted. Hematopviesis 26 days following transplant, and has been in complete remission for three years. It's evidenced that home produced monoclonal antibodies LS67-8 and LN93-2are both Safe and effective for marrow purging.
