小剂量rhTNF－α对HL－60细胞的诱导分化及增殖抑制作用的实验研究

摘　　要：采用细胞生长状态、细胞形态学变化、ＡＣＰ、α－ＮＡＥ酶活力和ＮＢＴ还原能力作为分化检测指标，研究了小剂量ｒｈＴＮＦ－α对ＨＬ－６０早幼粒白血病细胞株的诱导分化作用；同时采用细胞生长动力学，３Ｈ－ＴｄＲ掺入实验观察了ＴＮＦ诱导ＨＬ－６０细胞分化过程中对其生长增殖的抑制作用。结果发现：在ＴＮＦ５０～２００ｕ／ｍｌ的剂量下，对ＨＬ－６０细胞具有访导分化作用。使其向单核／巨噬细胞途径分化，其分化效率呈ＴＮＦ作用剂量和诱导时间依赖性；在ＨＬ－６０细胞分化同时尚伴有增殖抑制现象。We used the cell growth stages,cytomorphological changes,positive cell ratio of acid phosphatase(ACP),α-naphthylacetate(α-NAE), the abiliby to reduce NBT to study low-dose recombinanttumor necrosis factor-αon inducing differentiation of HL-60 cell line,Furthermore,the cell growthkinetics and 3H-TdR incorporation rate during TNF inducing HL-60 cell differentiation wereobserved. The results showed that TNF within the concentratin of 50-200u/ml can induce HL-60cell differentiation along the monocytic or marcrophage pathway. The differentiation ratio correletedwith TNP concentration and inducing time.The cell cytostatic effect appeared during differentiationprograms of HL-60 cells.
