摘  要:电镜观察不同禁水时间衰老大鼠下丘脑视上核加压素能神经元的超微结构变化。结果发现:禁水6、12h后,视上核加压素能神经元胞体增大,胞质中粗面内质网排列紧密且增多,高尔基复合体成熟面未成熟分泌颗粒及神经分泌颗粒增多;轴突中神经分泌颗粒增多不明显。而禁水24h后,神经元胞质内的神经分泌颗粒减少,轴突中的神经分泌颗粒聚集成膨大区域。禁水后,神经胶质细胞减少,突起回缩,使相邻的两神经元直接接触,突触结构重建增多。上述研究结果提示,衰老大鼠下丘脑视上核加压素能神经元在禁水时,其功能活动是增强的,胶质细胞突起回缩,有利于神经元之间的突触结构重建。Structural changes of the vasopressinergic neurons was less studied so far in aged rat during water deprivation. But changes of the vasopressin are closely related to aging processes. In order to study the relationship between the structural changes and the function of the vasopressinergic neurons,we selected aged male rats and divided them into a general group and three experimental groups of deprived water for 6, 12, 24 hours. We observed the ultrastructure of the vasopressinergic neurons during water deprivation in aged rat.Post-embedding PAP method was used by rabbit anti - AVP serum to the vasopressinergic neurons in the supraoptic nucleus. Under anesthesia ,rats were perfused via the ascending aorta with 4% paraformaldehyde -0. 25 % glularaldehyde in phosphate buffer. The hypothalamus was dissected out and cut into thin sections.At first,sections were incubated with rabbit anti-AVP,then goat anti-rabbit serum and peroxidase- anti peroxidase (PAP) were used ,and finally ,were stained with diaminobenzidine. The immunoreactive positions were flat embedded and dealt with the electron microscopic technique.The result showed:the vasopressinergic neuronal bodies became larger after deprivation of water for 6,12 hours. RER were arranged closely and their number was increased. The quantity of immature secretory granules in Golgi complex and neurosecretory granules (Nsg) were increased. But Nsg number in the axons did not change. After 24 hours of water deprivation Nsg in the vasopressinergic neuronal bodies were reduced ,and it existed in the axons and were accumulated in large groups. Moreover,in the neuropil processes of glial cell were retracted ,membrane apposition between adjacent neurons was formed and synapse formation were increased.It is concluded that during water deprivation the function of the vasopressinergic neurons in the hypothalamic supraoptic nucleus of aged rat is more active. The process retraction of glial cells has an effect on neuronal excitation.
