摘  要:首次用CFU-ALL极限稀释液体微培养法,对16例初诊,14例CR期ALL骨髓进行培养,并用8例正常,11例再生骨髓作为CFU-ALL阴性对照。结果显示:初诊B系ALL骨髓以重叠型集落伴CD(10+)细胞>20%为特点;初诊T-ALL集落也属重叠型(CD(2+)、CD(3+)、CD(7+)).而正常和再生骨髓虽形成集落,但为平展型或伴重叠型集落(CD(10+)<20%、以CD(15+)、CD(33+)细胞为主)。本文14例CR期ALL中,7例有重叠型集落(CD(10+)细胞>20%);临床随访1年半,5例骨髓相继复发,1例诊断为睾丸白血病。The cells of bone marrows from patients with childhood ALL(newly diagnosed:16 cases;in remission:14 cases)were culured undetr low O2 tension by means of micro-liquid CFU-ALL limited-dilution assay.Eight normal and eleven regenerating bone marrows were used as negative controls of CFU-ALL.The characters of colony cells were defined on the basis of colony morphologic appearance and colony cell-surface markers.The results showed:the newly diagnosed B-lineage ALL bone marrows yielded overlapping colonies with CD10+ cells>20%.The newly diagnosed T-ALL bone marrows also formed overlapping colonies(CD2+ CD5+ CD7+ cells).The normal and regenerating bone marrows also yielded colonies(flat or with overlapping colonies) under similar cultural conditions,but the number of CD10+ colony cells was low(<20%).The colonies mainly consisted of CD15+ CD33+ cells.On the basis of the characters of B lineage ALL colonies,the bone marrows of 7 patients with ALL in remission were detected to have overlapping colonies with CD10+ cells>20%,which was suspected to be the sign of residnal B-lineage ALL cells.These patients were followed up.The authors consider the assay will be valuable in studies on leukemia in vitro and detection of minimal residual disease.
