摘 要:目的 研究老年大鼠脑缺血 /再灌注 (I/R)微血管基底膜损伤的损点。方法 采用MCAO方法复制脑缺血模型 ,分为青年假手术组、老龄假手术组、青年模型组和老龄模型组 ,其中模型组又分脑缺血 3h和I/R 6h、12h、2 4h、3d、6d时间点。观察神经症状积分、梗死面积、病理变化、基底膜IV型胶原 (ColIV)和层连蛋白 (LN)。结果 随着再灌注时间延长 ,微血管基底膜损伤逐渐加重 ,I/R 2 4h、3d较显著。老龄假手术组ColIV和LN表达较青年假手术组增强。青年模型组 (I/R12h~ 6d)和老龄模型组 (I/R 6h~ 6d)ColIV和LN表达分别较青年假手术组和老龄假手术组减弱。老龄模型组神经积分 (I/R 2 4h~ 3d)的升高、梗死面积 (I/R 6h~ 6d)的增加、ColIV和LN(脑缺血 3h~I/R 2 4h)表达减弱较青年模型组显著。结论 I/R脑损伤和微血管基底膜破坏随着再灌注时间的延长而加重 ,老年较青年明显 ;微血管基底膜的增龄变化可能是其机制之一。Objective To study the characteristic of microvessel basement membrane during cerebral ischemia reperfusion in the aged rats.Methods We duplicated focal cerebral ischemia model with MCAO and divided the rat models into the young sham-operated group,the aged sham-operated group,the young model group and the aged model group, the latter two groups with ischemia 3 h and I/R 6 h,12 h,24 h,3 d,6 d points.The changes of the nervous dysfunction score,the area of cerebral infarction,the pathologic changes and the content of collagen type IV(Col IV) and laminin (LN) were observed.Results Along with the prolong of ischemia reperfusion time,the microvessel basement membrane lesion became more serious gradually peaking at I/R 24 h,3 d.The content of ColIV and LN of the aged sham-operated group were higher than those of the young sham-operated group.The content of Col IV and LN of the young model group(I/R 12 h~6 d) and the aged model groups (I/R 6 h~6 d) were lower respectively than those of the young sham-operated group and aged sham-operated group.The rise of the nervous dysfunction score(I/R 24 h~3 d),the increase of area of cerebral infarction(I/R6 h~6 d) and the decrease of content of Col IV and LN of the aged model group were more serious than those of the young model group.Conclusion Ischemia reperfusion brain lesion and microvessel basement membrane lesion were correlated to the ischemia reperfusion time while the lesion of the aged were more serious than that of the young.The microvessel basement membrane changes with age might be the one of the mechanisms.
