老年鼠胃窦黏膜与血浆生长抑素研究

摘　　要：目的研究老年期胃黏膜生长抑素含量和分布状况,探讨老化过程中生长抑素作为一种胃酸抑制因子存在的实际意义。方法选择不同年龄Wistar大鼠,雌雄随机。青年鼠(3月龄)20只,体重200～250g;老年鼠(24～30月龄)15只,体重400～450g。动物按规定月龄取血,分离血浆样本,提取胃黏膜组织,采用放射免疫分析法分别进行生长抑素含量测定。取胃窦新鲜组织,经冰冻切片进行生长抑素免疫组化染色。结果不同年龄组Wistar大鼠胃黏膜组织生长抑素具有不同的生理浓度,在老年鼠生长抑素含量呈高水平分布,与青年鼠比较有非常显著性差异(P<0.01)。生长抑素免疫组化染色显示,胃窦D细胞形状不规则,胞浆内布满生长抑素免疫复合物颗粒,老年鼠D细胞显色较深,其D细胞数量与免疫复合物颗粒高于青年鼠组。两组比较有显著性差异(P<0.05)。结论老年期胃黏膜D细胞数量增加,生长抑素分泌量增高,与其老年抑酸调节作用增强存在重要关系。Objective To study the topography and plasma level of somatostatin of rats in order to explore the effects of somatostatin in inhibiting acid secretion in the course of ageing. Methods Different Wistar rats were selected. The rats were divided into two groups.20 young rats(age 3 months) weight 200 ～ 250g, and 15 old rats (age 24 ～ 30 months) weight 400 ～ 450g. The somatostatin plasma level in rats was assayed by radioimmunoassay every month. At the meantime, the frozen section of the rats sinus ventriculi were obtained to study the topography of somatostatin by immunohistochemistry stain. Results The rats in different groups had different concentration of somatostatin with a high somatostatin level in old rats group. There was a significant difference between two groups(P < 0.01). The topography of somatostatin by immunochemistry stain showe that the rat gastric sinus D cell shape was irregular and full of somatostatin compound. The D cells had deeper colour. The quantity and concentration of D cells in old rats gastric sinus were more than that in young rats (age 3 months). The difference between two groups was singnificant (P < 0.05). Conclusions The quantity of D cells and the level of somatostatin in old rats gastric sinus mucosa are higher than that in young rats. These might associate with regulation disturbance of acid secretion in old rats.
