摘  要:采用试管静止富集培养及生态模拟富集培养,成功地从100份水样中分离到32株鞘细菌.利用Winogradsky液体试管法和摇瓶法进行氧化铁鞘细菌的快速筛选,从中筛选出1株氧化活性最高的菌株FC9901.对铁氧化酶特性进行了初步研究,该酶作用的最适pH和最适温度分别为7.5和30℃.金属离子Ca2+、Mg2+、Zn2+对氧化活性有激活和稳定作用;Cu2+、Hg2+、Al3+则抑制酶的活性;Fe2+、K+、Na+对酶活性影响不明显.32 strains of sheathed bacteria were successfully isolated from 100 water samples by two enrichment methods. Rapid screening of sheathed bacteria with Fe^(2+) oxidizing ability was done by Winogradsky liquid tube methods and flask methods. FC9901 was the best one which can produce oxidizing enzyme with the highest activity. Biochemical characteristic of the iron oxidizing enzyme were examined as follows, the optimum temperature and pH for oxidizing activity was 30℃ and 7.5. Ca^(2+) ?Mg^(2+) ?Zn^(2+) increased the oxidizing activity ,whereas Cu^(2+)?Hg^(2+) ?Al^(3+) caused inhibition .Fe^(2+)?K^(+) ?Na^(+) effected on the activity was not obvious.
