多巴胺D_4受体基因与注意缺陷多动障碍及其相关症状的关联分析

摘　　要：目的 探讨多巴胺D4受体 (dopamineD4receptor,DRD4 )基因 48bp可变重复序列 (variantnumbertandemrepeat,VNTR)多态性与注意缺陷多动障碍(Attention deficithyperactivitydisorder,ADHD)及其相关症状的关系。方法 取 139例ADHD患者及 115例正常对照,利用Achenbach父母用儿童行为调查表(ChildBehaviourChecklist,CBCL)来评定患者的临床症状;采用聚合酶链式反应(polymerasechainreac tion,PCR)、聚丙烯酰胺凝胶电泳结合银染技术,检测ADHD患者和对照组基因型和等位基因的频率。结果 患者组和对照组DRD4基因型和等位基因的频率与对照组相比无显著性差异 (P>0. 05)。DRD4基因的携带 5等位基因(DRD4 *5+ )组个体间焦虑 /抑郁(5. 1±4. 0)和内化性(12. 1±7. 6)两分量表分和非携带 5等位基因(DRD4 *5 )组个体间焦虑 /抑郁(2. 7±2. 7)和内化性 (7. 3±5. 4)两分量表分有显著性差异(Mann WhitneyZ=1. 982,P=0. 047;Z=2. 047, P=0. 041), DRD4 *5+基因型个体焦虑 /抑郁和内向性两行为分量表评分高。而其它分量表及行为总分无显著性差异。结论 本研究未发现DRD4基因 48bpVNTR多态性与ADHD存在关联,但DRD基因 48bpVNTR多态性与ADHD伴内化问题可能有关。ObjectiveTo investigate the relationship between 48bp variant number tandem repeat (VNTR) polymorphism in the third exon of dopamine D 4 receptor(DRD 4) gene and attention deficit hyperactivity disorder (ADHD). Methods We studied 139 patients with ADHD and 115 normal controls were studied. Patients' clinical symptoms were rated according to Child Behaviour Checklist (CBCL). The genotype and alleles in these patients and normal controls were examined with polymerase chain reaction(PCR) , non-denaturing polyacrylamide gel electrophoresis and silver staining . ResultsThere no significant differences in the frequencies of the genotypes and alleles of DRD 4 gene between ADHD and normal control(P>0.05). There were significant effects of DRD 4 genotypes for Anxious/ Depressed and Internalizing Problems of CBCL and subjects with 5 allele of DRD 4 gene (DRD 4*5 +)showed more Anxious/ Depressed and Internalizing Problems(Mann-Whitney Z=1.982,P= 0.047;Z=2.047,P=0.041).Conclusion The results suggested that ADHD and the DRD 4 gene was not significant association.but a effect of the DRD 4 on some ADHD symptomatology (Anxious/ Depressed and Internalizing Problems).
