摘 要:目的合成一条具有双亲功能的多肽,观察其生物活性。方法采用固相多肽合成技术制备具有双亲桥接功能多肽P19,一端含有多谷氨酸序列,与羟基磷灰石(HA)具有特殊的亲和力,另一端又具有精氨酸-甘氨酸-天门冬氨酸(RGD)三肽序列,通过整合素受体促进成骨细胞的黏附和增殖。观察P19对HA的亲和力和表面培养UMR106细胞的成骨活性。结果合成P19相对分子质量2215.26,纯度53.62%,与HA具有较高的亲和力,生理条件下不能被洗脱,同时P19能够促进UMR106细胞在HA表面的吸附、增殖,并提高碱性磷酸酶的活性。结论P19具有成骨活性,可复合HA修复骨缺损和用于HA表面喷涂假体的生物活性修饰成分。Objective To study the biological activity of the synthetic bi-affinity bridging peptides. Methods A peptide P19 with bi-affinity bridging function was prepared by solid phase polypeptide synthetic technique which this peptide contains a special hydroxyapatite-binding site(poly-glutamic acid sequence) and a cell-attachment site arginine-glycine-aspartic tripeptide sequence,RGD. The affinity of P19 to hydroxyapatite(HA) and the osteogenous activity of UMR106 were observed. Results The synthetic P19 could not be eluted from hydroxyapatite under physiological condition indicating there was high affinity between P19 and hydroxyapatite. The osteoblastic cell line UMR106 incubated with hydroxyapatite powder coated with P19 demonstrated that the cell adsorption to HA,proliferation and activity of alkaline phosphatase were promoted and elevated. Conclusion The results are showed that the P19 with osteogenous activity,can be used in the area of hydroxyapatite based materials for repairing bone defect and coating on HA prosthesis as a modification element with biological activity.
