摘 要:目的观察兔骨髓基质细胞在体外的培养条件及其成骨过程,为研制骨组织工程材料选择种子细胞奠定一定实验基础。方法取兔长管骨骨髓为材料进行体外培养,传代后改用条件培养基进行培养,通过倒置显微镜观察、碱性磷酸酶(alkalinephosphatase,ALP)染色、Ⅰ型胶原免疫组化染色等方法进行检测。结果传代细胞培养3、4d后融合成单层,培养12d细胞形成多层结构,并聚集成多个散在的黑色结节,细胞富含碱性磷酸酶活性,Ⅰ型胶原染色阳性,这与典型成骨细胞的生物学特性相似。结论体外培养的兔骨髓基质细胞具有成骨能力,可作为骨组织工程学研究的种子细胞。To observe the culture condition of the bone marrow stromal cells (BMSC) of rabbit in vitro and osteogensic process, and select the suitable seed-cells for bone tissue engineering. The little rabbits long bone marrow cells were cultured in vitro. The cells were passaged and subcultured in condition medium, and examined by phase contrast microscope, ALP activity and type Ⅰcollagen staining. The passaged cells were cultured in condition medium for 3, 4 d and lined up in one layer and gradually transformed into multilayer structure for 12 d and congregated into opaque clusters later, ALP and typeⅠcollagen staining showed positive reaction. The cells had the same morphological feature with the classical osteoblast cells. [Conclusions] The cultured rabbit BMSC in vitro possess osteogenic capability, and can be used as the seed-cells of the bone tissue engineeing.
